چکیده

Aim The propagation of HIV-1 infection represents a major problem of public health, notably in sub-Saharan Africa where about six out of ten infected adults are women. Among the new options of preventive therapy, antiretroviral drugs used as microbicides distributed via an intravaginal ring (IVR) constitutes an original solution. IVR is an interesting mode of drug delivery because it is a coit-independent device that can be left in the vagina for several months and can release regular amounts of microbicides. The aim of this work was to set up a silicone IVR able to deliver simultaneously several drugs with the precise objective of determining if the release of each drug is possible with the used silicones and of choosing the best formulation.

Development and in vitro/in vivo Evaluation of a Silastic Intravaginal Ring for Mifepristone Delivery

Preparation and in vitro/in vivo evaluation of mifepristone intravaginal ring formulations were investigated. In the present study, it is reported that a mifepristone intravaginal ring of reservoir design comprising of a mifepristone silicone elastomer core enclosed in a silicone layer. During the preparation of intravaginal ring solid dispersion method was employed which improved the release r...

In Vitro Investigation and Characterization of Silicone Matrix IVR Loaded with Levonorgestrel: A Propper Method for Hormon Replacement Therapy

Introduction: Among 3 to 30 percent of women of reproductive age, experience abnormal uterine bleeding (AUB). Vaginal rings are one of the systems of interest for the local delivery of hormones that due to their polymeric structure allow prolonged release of drug. In this study, the design and fabrication of matrix vaginal rings containing levonorgestrel (LNG) were investigated. Methods: In th...

Modified silicone elastomer vaginal gels for sustained release of antiretroviral HIV microbicides.

We previously reported nonaqueous silicone elastomer gels (SEGs) for sustained vaginal administration of the CCR5-targeted entry inhibitor maraviroc (MVC). Here, we describe chemically modified SEGs (h-SEGs) in which the hydrophobic cyclomethicone component was partially replaced with relatively hydrophilic silanol-terminated polydimethylsiloxanes (st-PDMS). MVC and emtricitabine (a nucleoside ...

Sustained delivery of commensal bacteria from pod-intravaginal rings.

Topical administration of live commensal bacteria to the vaginal tract holds significant potential as a cost-effective strategy for the treatment of sexually transmitted infections and the delivery of mucosal vaccines. Probiotic-releasing intravaginal rings (IVRs) embody significant theoretical advantages over traditional daily-dosage forms, such as sustained and controlled delivery leading to ...

Long-term, controlled release of the HIV microbicide TMC120 from silicone elastomer vaginal rings.

OBJECTIVES The feasibility of providing prolonged and controlled release of the experimental non-nucleoside reverse transcriptase inhibitor TMC120 from a silicone vaginal ring in quantities sufficient to maintain a vaginal concentration offering protection against heterosexual HIV transmission was investigated. METHODS Core-type, silicone elastomer vaginal rings containing TMC120 were manufac...
